About RecombiMAb anti-mouse OX40L (CD252, CD134L)

The RM134L-CP066 monoclonal antibody is a recombinant, murine chimeric version of the original RM134L antibody. The variable domain sequences are identical but the constant region sequences have been switched from rat IgG2b, κ to mouse IgG2a, κ for use in murine models. Species-matched chimeric antibodies exhibit regulated effector functions—including Fc receptor binding and complement activation—and cause less immunogenicity and formation of anti-drug antibodies (ADAs) than xenogenic antibodies in animal models. The highly controlled sequence and lack of genetic drift in recombinant antibodies provide more reliable and reproducible results over hybridoma derived antibodies. The RM134L monoclonal antibody reacts with mouse OX-40L (also known CD252, CD134L, TNFSF4). OX-40L is a 35 kDa member of the TNF superfamily that is expressed on activated B cells, dendritic cells, vascular endothelial cells, mast cells, and natural killer cells. OX40L is the ligand for OX-40 (CD134). OX-40 signaling regulates both CD4 and CD8 T cell clonal expansion, provides a costimulatory signal to an antigen-reacting naive T cells prolonging proliferation, augments the production of several cytokines including RANTES, IL-2, IL-3, and IFN-g and plays an important role in immunotolerance. In vivo treatment with the RM134L antibody has been shown to block the interaction between OX40L and OX40.
